Hi, Your information about "How to add POP-7 with a modified supply bottle?" was very interesting. Do you have any Idea about ABI 3500? I mean refilling pop-7 polymer. Best Regards. [Response, Aug. 2021: The 3500 no longer has a hard stop for when the polymer pouch would be consumed; thus, I suspect that one could pierce the pouch, add polymer, and seal the hole again... but have no 3500 instrument to test. The 3500 polymers also differ from those of the 3130; thus, I don't know if one can get the right polymer in a format that would allow for transferring polymer into an existing pouch.]

very useful. thanks a question, when the equipment arise the message Laser power probably is too dirty? [Response: Multiple issues might generate an error message about Laser power, but they would all likely require professional service to resolve the problem.]

This resource has been of the utmost help for me, as I was hired to be the "caretaker" of a 3130XL at a private DNA testing laboratory. I cannot even begin to stress how useful the information has been. Thank you so much!

Good Morning, I am, Dr. Summer Siddiqui, working as a postdoctoral fellow with Dr. Binhua Ling at TNPRC. I have some samples for sequencing and want to use DNA sequencing core facility. Can you send me basic information about pricing, sample submission procedure and length of time to get results? Thanks, Summer [Response: This facility is primarily for labs at the LSU Baton Rouge campus; however, if you email us through the Contact Us link, we can discuss your potential options further.]

Do you know how to prevent the piston rises up when you start the secuencer?. Thanks and regards. [Response: As far as I know, there is NO way to prevent the piston from initializing when the Sequencer is started. The “stop” command in the Manual Controls does not work; see "How to stop the piston?" in the 3130XL SACKS. If the sequencer goes offline (e.g., in a power failure), then one can only hope there is enough polymer in the supply bottle to keep air out of the supply tube. However, when I know that the sequencer will be turned off, I add enough polymer to the supply bottle to allow for the pump to initialize without sucking in any air.]

Thanks so very much for your excellent site. This is all such invaluable information and collected in one place. I am very interested in Capillary washing with water to try and extend the useful life - this is fantastic, thanks again.

Our lab have this machine and I have been doing the maintenance (and am almost the only user) for years. I wish I had found here earlier. This is very very very useful! Thank you! To me, whoever wrote up this protocol deserves a Nobel prize. [Response: Still no word from the Nobel committee...]

As requested, I am signing the guest book to let you know how useful I have found the 3130xl information. I haven't read all the information in full yet, but am thrilled to have found your site. The straightforward answers to questions are great, especially as I am struggling to run an instrument on my own with infrequent use - creates lots of problems! Particulary interested in the info about POP7 use and capiallary shelf life. Thank you!

This is great, thank you! Something I might be able to contribute: http://www.azcobiotech.com/reagents/NPPolymer.php inexpensive polymer, has worked very well for us. [Response: Options from other vendors can be less expensive and may work well. Do keep in mind that new spectrals are required if switching polymer vendors. Also, if your Service Agreement is through ThermoFisher (ABI), you will likely be required to put the instrument back on the standard reagents before an FSE will service the instrument.]

I suggest that the info on the timeline of obtaining a user ID be placed in a non- secure area....like maybe placed on the page where we initially request a user ID ! Also , LOVE the site...valuable , user-friendly information is provided . Impressive and greatly appreciated. Kudos! [Response: Good suggestion... done.]
